Before you can upload to Amazon, double check that your inventory template file is saved to your local hard drive.

Add Products via Upload
Go to Amazon Seller Central and hover over the INVENTORY menu. Click “Add Products via Upload”
Select Type of File to Upload
Find the section “Upload inventory files” and Select the type of file to upload. For most users this will be “Inventory Files for non-Media Categories” (such as Home & Garden, Office Products, Jewelry…)
Upload the Inventory File
Click “Choose File” and find the csv file you had previously saved to your hard drive. Amazon will process the file either begin listing your product, or generate an error report file for download.

Scroll down to the Inventory file upload process section. Download the report for more details.
Depending on the report, you may have to make modifications to your inventory template.
Edit your template for missing or required values.
Update: Amazon has made many upgrades since this post was published and edited.
